Engineered vitamin E-tethered non-immunogenic facial lipopeptide for developing improved siRNA based combination therapy against metastatic breast cancer

RNA interference (RNAi) is a promising method for treating highly metastatic breast cancer, but delivering functional siRNA to cells remains a major challenge. To address this, a research team has developed a vitamin E-tethered, protease-stabilized lipopeptide to transport siRNA effectively. This non-immunogenic, biocompatible transporter features an Arginine-Sarcosine-Arginine segment for stability and reduced aggregation, and α-tocopherol for better cell penetration. Their lipopeptide outperforms HiPerFect in long-term transfection and can transfect difficult primary cells like HUVEC.

Design Approach

The prominent hourglass symbolizes the gradual and sustained release of the payload from the nanoparticle. Inside the hourglass, intracellular events are depicted, while changes in the tumor microenvironment, such as the disruption of cancer-endothelial cell connections, are shown outside. It's not always necessary to visualize a cover strictly within the study's boundaries; what matters is conveying the essence in an attractive and innovative way.
